We have investigated the role of cytokine production and recognition in the age-related impairment of cell proliferative responses. Peripheral blood mononuclear cells (PBM) from a continuous population of 38 healthy subjects aged 22-97 years (20 healthy adults and 18 institutionalized elderly subjects selected according to the SE-NIEUR EURAGE protocol) were mitogen activated, and their ability to synthesize IL-2 and IL-4, to express the membrane-bound IL-2R, or to release its soluble form in culture supernatant was studied. Results suggest that reduction in the synthesis of IL-2 and IL-4 and in the release of the soluble form of IL-2R significantly contribute to the decrease in T cell proliferative responses observed with aging. However, T cell blasts stimulated with an excess of IL-2 display an increase in their ability to proliferate, suggesting that aging may not significantly alter cell proliferation if adequate amounts of cytokines are provided.
