A tetramethylammonium hydroxide (TMAH) based digestion method for the determination of total selenium content in wheat and mussel samples is proposed. The results achieved with the proposed digestion procedure were compared with those obtained after microwave oven acid mineralization. Graphite furnace-atomic absorption spectrometry (GF-AAS) was used for the final selenium determination. Efficiency of the proposed digestion method analyses was evaluated on a Wheat Flour Standard Reference Material (NIST 1567a) and on spiked samples. Hydride generation-atomic absorption spectrometric analyses showed the possibility of exploiting TMAH digestion also for speciation purposes.
