Cell DNA contents have been measured by flow cytometry in 35 patients with transitional bladder cancer (29 males, 6 females, age range 50-84 years). Eight patients had Ta tumours, 17 T1, 5 T2 and 5 T3. Grade II was the most frequently represented (26/35). Fifteen cases exhibited a diploid pattern (42.8%) and 20 (57.2%) were aneuploid, 13 monoclonal and 7 multiclonal. No significant difference in ploidy pattern distribution was observed in Ta-T1, G2 tumours, while infiltrating and G3 tumours were more frequently aneuploid. As far as percent of cells in S phase is concerned, similar values in tumour and healthy mucosa of the same patient (mean 15.85 ± 8.57 vs 12.12 ± 7.23, respectively), were observed while a lower percentage of S phase cells (mean 12.2 ± 7.61) was observed in diploid with respect to aneuploid tumours (mean 19.9 ± 8.12). In patients submitted to BCG therapy or M-VAC chemotherapy, pre- and post-treatment DNA index (DI) were evaluated. In 7 non-responding patients, post-treatment DI values were identical to those obtained before therapy, except in two cases in which one of the previous subpopulation was absent in the post-treatment sample. A longer period of follow-up is needed in order to assess the real prognostic significance of these observations.
